Plasmodium falciparum (Pf) has a solitary cytosolic nucleotide exchange factor that supports in the process of protein folding and degradation machinery. A molecular dynamics simulation was used to study the dynamics of HSP110 (for the first time in Pf) and its co-chaperone. Instead of being well conserved, PfHSP70\’s inter-domain dynamics change, but HSP110\’s dynamics can be seen in all of its domains when compared to its human homologs. To biochemically describe it; we made efforts to clone both the HSP110 and its distinct domains using diverse techniques. Nevertheless, we had challenges in purifying any of the proteins. HSP110 is a protein that has an extra insertion between its domains, which makes it a protein with a high degree of disorder. HSP110 and its co-chaperone can be found in both the nucleus and the cytoplasm. While at RNA (HSP110-70) and protein (HSP40) levels were raised. The interaction between PfHSP110 and HSP70-40 was verified using immunoprecipitation and through mass spectroscopy analysis reveals that HSP110 is involved in several cellular pathways, including translation, DNA binding, metabolic processes etc. It demonstrates the significance of HSP110 in the survival of Pf under various stress conditions and when exposed to drugs.
